Columbia Pictures decided to limit the scope to two villains because it felt that too many villains weakened the last film - Some argue, however box office reciepts would suggest otherwise. There may be one villain, but two is the limit.
A lot of speculation is circling around the Lizard, whose alter-ego, Dr. Curt Connors, was shown in the last two movies, so there is hope for this one. Although we're not certain who would be the second villain or if there will be only one, however, you can bet that Dylan Baker will return to portray Dr. Connors. Raimi has also expressed some interest in including the vampiric Morbius as a villain, though this is unlikely because Morbius is a less popular character. There is also some speculation that Carnage will be the villain, and the movie will be based around the famous Maximum Carnage storyline, as well as Venom possibly making a return.
Spider-Man, obviously, and it's been suggested by interviews with Sam Raimi that Mary Jane Watson is returning as well. Besides those two, it's unclear who else is returning. JK Simmons also expressed interest in returning as J. Jonah Jameson.Recently, it has been reported that Dylan Baker will reprise his role as Peter's physics professor, Dr. Curt Connors, leading some to believe that he will also be portraying Connors' alter-ego, The Lizard.Also, Bruce Campbell, who has played an apparently separate role in each Spider-Man movie, has been confirmed to return. His role in this movie is currently unknown.
